April is characterized by increasing intensity of sunlight in the Northern Hemisphere due to the sun rising higher above the horizon, which is not only inspiring for our spirits here in the northern US, but it also inspired our latest product showcase for this newsletter.

PRODUCT HIGHLIGHTS

Quantum™ MESF Kits

Quantum™ MESF Kits contain one blank population and a series of either 4 or 5 microsphere populations with increasing fluorescence intensity, like the April sun’s increasing intensity. Each of these populations is assigned a Molecules of Equivalent Soluble Fluorochrome (MESF) value that corresponds to the fluorescence intensity of the pure fluorochrome molecule in solution.

To use:

  1. Analyze each Quantum™ MESF population on your flow cytometer on the same day with the same settings as your labeled cells.
  2. Record the median channel values.
  3. Enter these values into QuickCal®, our regression curve generating template. QuickCal® is provided free-of-charge with your purchase.
  4. Use QuickCal® to convert the channel value for your cell into an MESF value so you can compare instruments and correct for any environmental influences.

PRODUCT FAQs

Which kit do I need? The fluorophore used to stain your cell must match the fluorophore in our MESF kits.

What can I do if my cells are not stained with the same fluorophore?
If a suitable antibody labeled with a matching fluorophore cannot be found, then our Quantum™ Simply Cellular® Kits could be used. These have one blank and 4 populations of beads with increasing numbers of human, mouse, or rat capture antibodies. For more information, see PDS 818, Quantum™ Simply Cellular® & Quantum™ MESF Tips and Techniques.

My experimental sample does not fall within the linear range of my Quantum™ MESF Kit, can I extrapolate my calibration curve?
Yes, extrapolation is used for many applications. A search for relevant published literature supports the use of extrapolated values.
